Mais les avanc\u00e9es technologiques r\u00e9centes ont rendu <strong><a href=\"https:\/\/leyton.com\/ca\/insights\/articles\/rsde-dans-lindustrie-pharmaceutique\/\">la recherche et le d\u00e9veloppement pharmaceutiques<\/a><\/strong> plus accessibles et efficaces.<\/p>\n<p>Pour \u00e9tudier chaque maladie, les scientifiques doivent examiner les <strong>r\u00e9cepteurs<\/strong>, les <strong>enzymes<\/strong>, les <strong>prot\u00e9ines <\/strong>et les <strong>g\u00e8nes <\/strong>associ\u00e9s. C&#8217;est pourquoi l&#8217;identification de la cible est la premi\u00e8re \u00e9tape du processus.<\/p>\n<p>Une bonne cible m\u00e9dicamenteuse doit \u00eatre pertinente par rapport au <strong>ph\u00e9notype <\/strong>de la maladie. Mais doit aussi \u00eatre adapt\u00e9e \u00e0 une <strong>modulation th\u00e9rapeutique<\/strong> (\u00ab <strong>pharmacopotentialit\u00e9<\/strong>\u00bb).<\/p>\n<p>Les chercheurs doivent \u00e9galement s&#8217;assurer que le m\u00e9dicament pr\u00e9sente un b\u00e9n\u00e9fice th\u00e9rapeutique dans <strong>une marge de s\u00e9curit\u00e9 acceptable<\/strong>.<\/p>\n<p>Un scientifique peut constater qu&#8217;un m\u00e9dicament affecte la cible pr\u00e9vue. Mais devra encore \u00e9valuer le mode d&#8217;action pour garantir la s\u00e9curit\u00e9 du patient en utilisant des mod\u00e8les cellulaires, des mod\u00e8les animaux et, finalement, des essais cliniques.<\/p>\n<h2 class=\"wp-block-heading\">Comment l&#8217;IA est-elle utilis\u00e9e dans le d\u00e9veloppement de m\u00e9dicaments ?<\/h2>\n<p>L&#8217;analyse de <strong>donn\u00e9es biologiques multidimensionnelles<\/strong> n\u00e9cessite des m\u00e9thodes efficaces pour produire des pr\u00e9dictions pr\u00e9cises pour l&#8217;identification des cibles.<\/p>\n<p>L&#8217;<strong><a href=\"https:\/\/leyton.com\/ca\/insights\/articles\/comment-les-projets-dia-changent-la-donne\/\">IA<\/a><\/strong> est une<strong> technologie robuste<\/strong> utilis\u00e9e pour analyser les <strong>donn\u00e9es multi-omiques en rapide augmentation<\/strong> dans <strong>l&#8217;identification de cibles th\u00e9rapeutiques potentielles<\/strong>.<\/p>\n<h3 class=\"wp-block-heading\">Priorisation des cibles<\/h3>\n<p>La nomination de nouvelles cibles m\u00e9dicamenteuses et leur validation peuvent \u00eatre co\u00fbteuses et chronophages. Historiquement utilis\u00e9 pour reconna\u00eetre l&#8217;\u00e9criture manuscrite, d\u00e9tecter les visages ou identifier un locuteur.<\/p>\n<p><strong><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC4143549\/\">Jeon et al. (2014)<\/a><\/strong> ont construit un <strong>classificateur SVM<\/strong> utilisant des caract\u00e9ristiques provenant de divers types de donn\u00e9es (nombre de copies d&#8217;ADN, expression de l&#8217;ARN messager, occurrence de mutations et interactions prot\u00e9ine-prot\u00e9ine) pour prioriser les <strong>cibles m\u00e9dicamenteuses<\/strong> sp\u00e9cifiques aux cancers du pancr\u00e9as, du sein et de l&#8217;ovaire, afin d&#8217;<strong>identifier <\/strong>et de <strong>prioriser de nouvelles cibles m\u00e9dicamenteuses contre le cancer<\/strong>.<\/p>\n<p>L&#8217;<strong>algorithme SVM <\/strong>est puissant, ils pr\u00e9voient de d\u00e9velopper de <strong>nouvelles fonctions noyau<\/strong> pour aider \u00e0 la d\u00e9couverte de nouvelles cibles pour des <strong>cancers h\u00e9t\u00e9rog\u00e8nes<\/strong>. Tels que les cancers du sein triple n\u00e9gatif (TNBC) et les sarcomes des tissus mous (STS).<\/p>\n<h3 class=\"wp-block-heading\">Pharmacopotentialit\u00e9<\/h3>\n<p>La probabilit\u00e9 d&#8217;influencer une cible sp\u00e9cifique avec un <strong>m\u00e9dicament \u00e0 petite mol\u00e9cule<\/strong> d\u00e9pend de la capacit\u00e9 de la cible \u00e0 se lier \u00e0 de petites mol\u00e9cules en fonction de ses caract\u00e9ristiques biophysiques.<\/p>\n<p>Les entreprises de <strong>d\u00e9couverte de m\u00e9dicaments par Intelligence Artificielle<\/strong> ont abord\u00e9 la <strong>pharmacopotentialit\u00e9 <\/strong>en entra\u00eenant des mod\u00e8les pour l&#8217;estimer en utilisant diff\u00e9rentes propri\u00e9t\u00e9s. Notamment les caract\u00e9ristiques g\u00e9om\u00e9triques, structurelles et physico-chimiques des cavit\u00e9s de liaison aux m\u00e9dicaments et des cavit\u00e9s non liantes sur les prot\u00e9ines.<\/p>\n<p>Cela a aid\u00e9 les chercheurs \u00e0 d\u00e9couvrir que les attributs les plus critiques pour estimer la pharmacopotentialit\u00e9 sont la taille et la forme des cavit\u00e9s de surface de la prot\u00e9ine.<\/p>\n<p>Par exemple, <strong><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC3045802\/\">Costa et al. (2010)<\/a><\/strong> ont d\u00e9velopp\u00e9 un m\u00e9ta-classificateur bas\u00e9 sur un arbre de d\u00e9cision en s&#8217;entra\u00eenant sur des attributs tels que les caract\u00e9ristiques topologiques du r\u00e9seau, les profils d&#8217;expression tissulaire et la localisation subcellulaire pour chaque g\u00e8ne pharmacopotentiel et non pharmacopotentiel.<\/p>\n<p>Il a <strong>correctement identifi\u00e9 65 %<\/strong> des <strong>g\u00e8nes morbides connus<\/strong> avec une <strong>pr\u00e9cision de 66 %<\/strong> et a <strong>correctement identifi\u00e9 78 % <\/strong>des <strong>g\u00e8nes pharmacopotentiel connus <\/strong>avec une <strong>pr\u00e9cision de 75 %<\/strong>. Ce domaine n\u00e9cessite encore de l&#8217;innovation.<\/p>\n<h2 class=\"wp-block-heading\">Pr\u00e9dictions ADME-T (Absorption, Distribution, M\u00e9tabolisme, Excr\u00e9tion et Toxicit\u00e9)<\/h2>\n<p>Les propri\u00e9t\u00e9s <strong>ADME-T<\/strong> sont responsables d&#8217;environ la moiti\u00e9 de tous les \u00e9checs cliniques, c&#8217;est pourquoi il est essentiel d&#8217;am\u00e9liorer ce processus avec des applications d&#8217;<strong>IA<\/strong>.<\/p>\n<p>La plupart des mod\u00e8les de pr\u00e9diction tentent d&#8217;\u00e9tablir une relation directe entre les descripteurs mol\u00e9culaires et les propri\u00e9t\u00e9s <strong>ADME<\/strong>.<\/p>\n<p>L&#8217;introduction des <strong>r\u00e9seaux de capsules<\/strong>, un syst\u00e8me d&#8217;apprentissage automatique qui est un type de <strong>r\u00e9seau neuronal profond (DNN)<\/strong> pouvant \u00eatre utilis\u00e9 pour mod\u00e9liser des relations hi\u00e9rarchiques, a am\u00e9lior\u00e9 la<strong> pr\u00e9diction ADME-T<\/strong>.<\/p>\n<p>Par exemple, pour pr\u00e9dire la cardiotoxicit\u00e9 des m\u00e9dicaments, <strong><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC6997788\/\">Wang et al.
